I am trying to whole raising your hand if you want to talk, and she has to wait a minute til someone is done talking. She is going to Kindergarten in the fall so I figured if I start now with her raising her hand at home when she wants to talk it would get her the idea of getting used to it at school.

Ok, she's old enough to understand manners. You do not respond to her if she's interrupting. If she tugs, pulls, etc, you gently redirect her without acknowledging her.
As Louise posted, hold up your hand (the "stop" gesture) and continue your conversation.
Mine were the same, all kids are. When you finish your conversation, you then turn to the child, and say "now that you are not interrupting me, what's going on?"
You also need to teach how to politely disturb a conversation if it is important. Because, if little brother has cut himself, and she's trying to tell you that, she needs to have a way to do so.

How to stop this is to just show her the palm of your hand and continue to have your conversation with the other person. Do not give her eye contact. When your chat is over put your hand down and give her eye contact and allow her to speak. Then tell her again that it is rude to interupt and she has to wait until that person has finished. Praise her for being quiet when you raise your palm. She will learn very quickly to stop.
