Call the grand parents and talk to them in a civil manner. If that doesn't work, you can certainly get a new birth certificate. That will cost you about 15 bucks. Get ahold of your ex and find out exactly what is going on and pursue it from there. Why does your ex have custody and not you? That is a starting point.

My son is 7 yr no court give me letter saying he had custody . Could he have done it without me knowing?? I don't have the birth card his grantpartent has it

Do you have any court ordered custody papers? The only way (and it's fightable in court) that your ex's mom can keep your son is if your ex has 100% custody... in which case I would speak to a lawyer before doing anything. If you have ANY custody whatsoever... take the court order (or birth certificate if there is no court order) and go to the police and go get your son.
Is the ex planning on being gone for a long time... or is this just like a couple day trip?
Just reread your title and I'm assuming 'know has custody' means that no one has custody? In that case, grab his birth certificate and go to grandma's house. If she won't let you take the boy... call the police.
How old is your son though and does he WANT to come to you? If he's in his teens and wants to stay at grandma's... that may change what I think you should do.
